Background: The Platelet-derived growth factor (PDGF) family, characterized as a mitogen for fibroblasts and cells of mesenchymal origin in the late seventies, consists of five isoforms that are homodimers of A, B, C, and D polypeptide chains (forming PDGF-AA, -BB, -CC, and -DD, and a heterodimer PDGF-AB). The PDGF isoforms exert their cellular effects by binding to α and β‐ tyrosine kinase PDGF receptors. The PDGFR belong to the receptor-tyrosine kinase family, more precisely to the type III group, which also includes c-KIT, FLT3 and the macrophage-colony-stimulating factor receptor. PDGFRα binds to all ligands but PDGF-D, the PDGFRβ binds to PDGF-B and -D only. The two PDGF receptors are both consisting of five extracellular immunoglobulin-like domains, a single-spanning transmembrane domain and an intracellular split kinase domain, which is divided in two lobes connected by a flexible polypeptide linker, the kinase insert. Ligand induced receptor dimerization allows autophosphorylation in trans between the receptor subunits in the dimer. PDGFR-α and PDGFR-β engage several well-characterized signaling pathways e.g. Ras-MAPK, PI3K, and PLC-γ—which are known to be involved in multiple cellular and developmental responses. PDGF receptors interact also with integrins, which enhances cell proliferation, migration, and survival.
