Background
Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMPs are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The enzyme encoded by this gene degrades type IV and V collagens. Studies in rhesus monkeys suggest that the enzyme is involved in IL-8-induced mobilization of hematopoietic progenitor cells from bone marrow, and murine studies suggest a role in tumor-associated tissue remodeling. Thrombospondins, intervertebral disc proteins, regulate the effective levels of matrix metalloproteinases (MMPs) 2 and 9, which are key effectors of ECM remodeling. Matrix metalloproteinase-1 (MMP-1) also known as interstitial collagenase and fibroblast collagenase. MMP­1 is expressed by fibroblasts, keratinocytes, endothelial cells, monocytes and macrophages. MMP-1 breaks down the interstitial collagens, types I, II, and III. MMP­1 can degrade a broad range of substrates including types I, II, III, VII, VIII, and X collagens as well as casein, gelatin, myelin basic protein, L­Selectin, pro­TNF, IL­1β, IGF­BP3, IGF­BP5, pro MMP­2 and pro MMP­9.

Recombinant Human MMP1 Protein (rhMMP1) Phe 20 - Asn 469 (Accession # NP_002412.1) was produced in human 293 cells (HEK293).

Molecular Characterization
rhMMP1, fused with 6×His tag at the N-terminus, has a calculated MW of 52.7 kDa. The predicted N-terminus is Phe 20. DTT-reduced Protein migrates as 52-55 kDa.

Endotoxin
Less than 1.0 EU per μg of the rhMMP1 by the LAL method.

Purity
>95% as determined by SDS-PAGE.

Formulation
Lyophilized from 0.22 μm filtered solution in 20 mM MES, 150 mM NaCl, 10 mM CaCl2, 0.05% Brij-35, pH6.0. Normally trehalose is added as protectant before lyophilization.

Storage
Avoid repeated freeze-thaw cycles.

In lyophilized state for 1 year (4°C-8°C); After reconstitution under sterile conditions for 1 month (4°C-8°C) or 3 months (-20°C to -70°C).

Enzymatic Activity
Pre-activation is required for enzymatic assays. Please dilute Human MMP-1 to 100 µg/mL in TCNB buffer (50 mM Tris, 10 mM CaCl2, 150 mM NaCl, 0.05% Brij-35 (w/v), pH 7.5), and then add p-aminophenylmercuric acetate to a final concentration of 1 mM. Please keep the enzyme with APMA for 1 hour at 37°C.
100mM APMA stock solution should be prepared in DMSO. Please avoid adding high concentration APMA solution (>20mM) directly into the reaction as it tends to precipitate. A pre-dilution step is highly recommended.
