Background: The vaccinia-related kinase (VRK) protein family is a family of serine/threonine kinases composed of three proteins in mammals, VRK1, VRK2 and VRK3. The protein kinase VRK1 is mostly a chromatin kinase and regulates histones, but is also present in other complexes of nucleic acids, including RNA, and nucleoproteins. VRK1 is activated in response to mitogenic signals and regulates several transcription factors, including p53, ATF2 and c-Jun and histones and its covalent modifications. VRK1 is involved in regulation of cell proliferation and cell cycle regulation, e.g. induces the G1/S transition in cell cycle by promoting the expression of cyclin D1 in the G1/S phases by phosphorylating cAMP response element (CRE)-binding protein (CREB).
