Background: Testis-specific serine/threonine kinases (TSSKs) are a family of kinases in the CAM kinase family which are expressed in mammalian testis and are involved in spermiogenesis and fertilization. TSK2 has been localized to various regions of sperm, but mainly in the centrioles of post-meiotic spermatids during spermiogenesis, and it has been postulated that TSK2 has an important role in germ cell differentiation and/or sperm function. Human TSK2 phosphorylates the N-terminal region of TSKS, an endogenous substrate, and displays robust autophosphorylation. Single nucleotide polymorphisms of the TSSK2 gene are associated with impairment of spermatogenesis in idiopathically infertile males.
