Background: STK16 (serine/threonine kinase 16, also known as Krct, PKL12, MPSK1, and TSF-1, TGF-beta stimulated factor 1) is a myristoylated and palmitoylated serine/threonine-protein kinase and a member of the NAK (Numb Associated Kinase) subfamily. The kinase is ubiquitously expressed with highest expression observed in the liver, testis, and kidney and conserved among all of the eukaryotes. STK16 localizes to the Golgi complex throughout the cell cycle and plays important roles in Golgi structure regulation. Thus, STK16 knockdown or kinase inhibition not only delays mitotic entry and prolongs mitosis, but also causes prometaphase and cytokinesis arrest. Furthermore, TSF1/STK16 interact with MAL2 protein, GlcNAcK kinase, and DRG1 protein, is involved in the TGF-β signaling pathway and in regulation of VEGF expression.
