Background: PDZ binding kinase/lymphokine-activated killer T-cell –originated protein kinase (PBK/TOPK) is a 322 amino acid mitotic protein kinase and a member of the mitogen-activated protein kinase kinase (MAPKK) family. It is reported that PBK/TOPK can phosphorylate p38 and is suggested to be a novel MEK3/6-related MAPKK. PBK/TOPK is also involved in the cytokinetic function, DNA damage and repair and plays an important role in spermatogenesis. In cell cycle regulation TOPK plays a major role by acting as a substrate for cdc2/cyclin B (CDK1). Furthermore, PBK/TOPK is also expressed in hematologic tumors such as leukemia, lymphoma, and myeloma, and its expression was correlated with malignant potential of these tumors.
