Background: The c-Jun N-terminal kinases JNK / SAPK are members of the MAP kinase family acting as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. Complete activation of JNKs requires dual phosphorylation within the activation loop by MKK4 and / or MKK7. Activated JNK2 can in turn phosphorylate transcription factors such as c-Jun and ATF2, which participate in the activation and formation of the AP-1 complex and anti-apoptotic Bcl-2 family members as well as pro-apoptotic BAD proteins. In contrast, the inactive JNKs have been shown to cause degradation of several transcription factors (c-Jun, ATF2, p53). The expression level and the activity of JNKs is altered in various cancer types. Based on several experiments inhibition of JNK2 is associated with an reduced tumor growth, whereas blocking of JNK1 seems to results in an increased risk of tumorigenesis
