The Ser/Thr-specific phosphatases can be divided into two major families: the PPP family, which includes PP1, PP2A, PP2B/calcineurin and the PPM family composed of PP2C. The PPM group has a strict requirement for divalent metal ions (Mg2+, Mn2+) and is insensitive to known PPP inhibitors. Mammalian PP2C was initially found to exist as two highly conserved isoforms (α and β) which show approximately 70% sequence homology. PP2Cα can be stimulated 10�15-fold by certain lipophilic compounds. By unsaturated fatty acids from lipoproteins activated PP2Cα is involved in triggering apoptosis by dephosphorylation of the pro-apoptotic protein BAD. PP2Cα also upregulates the expression and the activity of p53 and reduces the activities of CDK2 and CDK6 as well as MAPK.
