Background: PKCη (PKC eta) is a member of the novel PKC isozymes that regulates cell proliferation, differentiation, secretion and apoptosis. The calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase is primarily expressed in epithelial cells. It was reported that PKC eta distributes in the ER, the Golgi apparatus, and the nuclear envelope in epithelial cells and is constitutively present in lipid droplet (LD), an organelle made of a lipid ester core and a surface phospholipid monolayer.
The regulation of PKC eta is unique compared to other PKC isozymes. Whereas phorbol esters are potent activators of conventional and novel PKCs a persistent treatment leads to the downregulation causing termination of PKC signaling. In contrast PKCη is the only phorbol ester-sensitive PKC isozyme that resists downregulation. Furthermore, PKC eta is activated by skin epidermis uique lipids like cholesterolsulfate and suifatide, sulfated metabolites of cholesterol and cerebroside.
PKCη binds and activates the tyrosine kinase FYN in keratinocytes, which in turn blocks epidermal growth factor receptor (EGFR) signaling. In addition, PKC eta provides negative control on AKT pathway since in in MCF-7 breast adenocarcinoma cancer cells the IGF-I induced phosphorylation on Ser473 of AKT was inhibited by the PKC eta.
