Antibody-Antigen Interactions

Antibodies are helpful analytical, diagnostic and therapeutic tools for a number of different applications. Classical methods for the kinetic characterisation of antigen - antibody interactions like ELISA only provide steady state data at one defined time point not considering the rate for complex formation and complex dissociation. BIA technology monitors the binding kinetics between antigens and antibodies in real time. The binding curves obtained at different concentrations contain information about the rates for association and dissociation, about the stochiometry of binding and thus about the binding activity and quality of different fractions within a protein solution, but also about heterogeneity, biphasic binding behaviour and avidity effects.

Furthers applications of the BIA technology in antibody characterisation are :

  • Qualitative ranking of crude antibody samples for comparing off rates
  • Quantitative kinetic characterisation yielding kass, kdiss and KD values
  • Optimisation of experimental conditions for antibody based assays
  • Pair-wise epitope-mapping for comparing binding epitopes of antibodies
  • Testing cross reactivity of antibodies against different antigens and species
  • Quality control of recombinant antigen preparations by binding assays
  • Testing immunoreactivity of pharmaceutical compounds in animal samples
  • Quantitative determination of antibody concentrations in serum samples
  • Identification of antibody classes and subtypes (IgG, IgE, IgM, IgG1,...)
