PDGF, a growth factor originally purified from platelets, is a family of disulfide-bonded homo- and heterodimers which are encoded by four genes PDGF-A, PDGF-B, PDGF-C, and PDGF-D. Each PDGF chain contains a highly conserved growth factor core domain, consisting of a set of cysteine residues. The four dimeric isoforms, PDGF-AA, PDGFAB, PDGF-BB and PDGF-CC can bind to and active PDGFRα, while PDGF-BB and PDGF-DD can specifically bind to and active PDGFRβ. Receptor activation leads to further signal propagation involving e.g. the Ras-MAPK, PI3K, and PLC-γ pathway.
Synonyms: Glioma-derived growth factor, GDGF, Osteosarcoma-derived Growth Factor, ODGF, SIS, SSV, PDGF2, c-sis, FLJ12858, PDGF-BB, PDGF B-chain, Platelet-derived growth factor beta polypeptide, Becaplermin
Protein informations: Platelet-Derived Growth Factor BB, human recombinant, is a homodimeric, non-glycosylated, polypeptide chain containing 109 amino acids and having a molecular mass of 24.3 kDa.
PDGF-BB is purified by proprietary chromatographic techniques.
Purity: > 95% by SDS PAGE
Biological activity: The ED50, calculated by the dose-dependent proliferation of BALB/c 3T3 indicator cells (measured by 3H-thymidine uptake) is < 1 ng/ml, corresponding to a specific activity of 1.000.000 U/mg.
