PTP-1B is part of the non transmembrane subfamily of classical protein tyrosine phosphatases. The protein consists of an N-terminal PTP-domain followed by two tandem proline-rich sequences and a C-terminal hydrophobic stretch, targeting the enzyme to the endoplasmatic reticulum. The catalytic PTP-domain contains a cysteine residue within its active site that mediates the nucleophilic attack on substrate phosphotyrosine residues. PTP1B negatively regulates several signalling pathways by dephosphorylating EGFR and PDGFR and has a prominent role in IR signalling and glucose homeostasis. Dysregulation of PTP1-B is associated with insulin resistance and diabetes and obesity.
