Background: ZAP70 (Zeta-chain associated protein kinase) is a 70 kDa member of the Syk family kinase predominantly involved in T cell receptor (TCR) signaling. However, recent findings have revealed involvement of ZAP70 in B cell receptor (BCR) signaling especially in leukemic and primary B cells too. The cytosolic protein kinase is characterized by the presence of tandem Src-homology 2 (SH2) domains preceding a highly conserved catalytic domain. Following TCR stimulation ZAP70 is recruited to the plasma membrane mediated by binding of the two SH2 domains of ZAP70 to the doubly phosphorylated immunoreceptor tyrosine-based activation motif (ITAMs) of the TCR.  This translocation process enables ZAP70 to be activated by Lck mediated phosphorylation on numerous tyrosine residues, but also contributes to relieve an autoinhibited conformation of the kinase. Phosphorylated ZAP70 in turn activates intermediates such as linker for activation (LAT) and SLP-76 leading to calcium mobilization and activation of downstream signaling pathways (MAPK and PI3 kinase).  ZAP70 controls cytoskeleton modifications, adhesion and mobility of T lymphocytes and is involved in human thymocyte development. Defects in ZAP70 signaling have been associated with autoimmunity  and cancer.
