Background: Mammalian Sterile20-like kinase 1 (Mst1) is a serine/threonine kinase which represents a member of the germinal center kinases (GCKs), a family of ‘Sterile 20 (STE20) like kinases’, which participate in both the determination of cell fate and the regulation of cell functions. MST1 and MST2 are mammalian homologs of drosophila ‘Hpo’. The MST1 /STEK4 contains an N-terminal catalytic domain, followed by an autoinhibitory segment and a coiled-coil SARAH domain that mediates hetero- and homo-dimerisation. MST1 has been implicated in T-cell biology. Thus, cross-talk between MST1 and Akt appears able to antagonize Akt1 activity which is involved in T-cell co-stimulation and the regulation of NF-κB-dependent gene transcription. MST1/2 in conjunction with an adaptor protein Salvador phosphorylates and activates LATS1/2 kinases which finally results in an inhibition of cell proliferation and promotion of apoptosis.
