Background: The mammalian Ste20 family consists of two structurally distinct subfamilies, including the P21-activated kinase family (PAK) and the germinal center kinase (GCK) family. MST1 and MST2 are members of the GCK II subfamily. MST4 belong as well as MST3 to the GCK III subfamily and is highly expressed in placenta, thymus, and peripheral blood leukocytes. MST4 is composed of an N-terminal kinase domain and a C-terminal regulatory domain which mediates homodimerization, and influences kinase activation. MST4 may activate the extracellular signal-regulated kinase (ERK) signaling pathway and acts downstream of liver kinase B1 (LKB1), a heterotrimeric protein consisting of the scaffold protein MO25 and the catalytically inactive pseudokinase STRAD (STE20-related adaptor). Mst4 translocates from the Golgi to the subapical membrane compartment upon activation of Lkb1, phosphorylates the regulatory T567 residue of Ezrin and induce brush border formation.
